Specimen Collection Details
Collection: One 10 mL green top tube (heparin). Also acceptable: One 4 mL lavender top tube (EDTA).

NOTE: Patient should fast 12-14 hours (water only) and abstain from alcohol 24 hours prior to collection.

Handling: Immediately place on wet ice. Transfer to a 12 mL graduated centrifuge tube and centrifuge 10 minutes at 2000 rpm. Record volume of packed cells. Discard supernatant plasma. Wash erythrocytes 2 times by resuspension in 5.0 mL cold 0.9% saline, discarding supernatant after each washing. Resuspend packed cells to original volume with 0.9% saline. Invert gently to mix. Record volume of packed cells and total volume of erythrocyte suspension on requisition. Freeze.
Standard Volume: 5 mL RBC resuspension
Minimum Volume: 1 mL RBC resuspension
Transport: Frozen.
Comments: Useful in confirming a diagnosis of aminolevulinic acid dehydratase deficiency porphyria (ADP).
NOTE: This assay re-activates ALAD that has been inhibited by lead, and therefore it is not useful in assessment of lead intoxication.

Rejection Criteria: Thawed specimen; hemolysis; frozen whole blood.
