BLOOD: One 4 mL lavender top (EDTA), 4 mL green top (sodium heparin) or 6 mL yellow top tube (ACD); BODY FLUID: At least 5 mL fluid preferred. For fluids with low cell counts, larger volumes will improve results.
Handling:
BONE MARROW, BLOOD, or BODY FLUIDS: Mix with equal volumes of RPMI media, available from Client Services. Submit to laboratory within 24 hours.
Comments:
DNA ploidy analysis provides correlative information of use in the staging of tumors. Ploidy analysis provides indication of whether a tumor has a normal component of chromosomes (diploid) or an abnormal number (aneuploid). The percentage of cells which are in active growth (S-phase) can also be enumerated. Generally, tumors which are aneuploid and have high S-phase percentages behave more aggressively.
Rejection Criteria:
Insufficient cells for analysis; excessive necrosis and debris.
