Specimen Collection Details
Collection: One 7.5 mL serum separator tube (SST).
Handling: Allow to clot, centrifuge and immediately separate serum. Label specimen as ‘acute’ or convalescent.’
Stability: 48 hours ambient, 14 days refrigerated, or 1 year frozen. Avoid repeated freeze/thaw cycles.
Standard Volume: 1 mL serum.
Minimum Volume: 100 µL serum.
Transport: Refrigerated.
Rejection Criteria: Severly lipemic, contaminated, heat-inactivated, or hemolyzed specimen.

Interpretive Data IV=Index Value: Optical Density (OD) of the patient serum divided by the OD of the cutoff value.
≤ 0.79 IV Negative: No significant level of detectable E. histolytica IgG antibody.
0.80-1.19 IV Equivocal: Repeat testing in 10-14 days may be helpful.
≥ 1.20 IV Positive: IgG antibody to E. histolytica detected, which may indicate a current or recent infection.
Seroconversion between acute and convalescent sera is considered strong evidence of recent infection. The best evidence for infection is a significant change on two appropriately timed specimens where both tests are done in the same laboratory at the same time.

In the case of extraintestinal complications, a postivie antibody can indicate amebiasis even though stool findings are negative.
