Specimen Collection Details
Collection:
One 10 mL green top tube (sodium heparin). Patient must be fasting 12-14 hours. Water only may be taken as needed; no other liquids are allowed. No alcohol for 24 hours prior to collection.
Handling:
Place on wet ice immediately. Transfer to a 12 mL graduated centrifuge tube. Centrifuge 10 minutes at 2000 rpm. Record volume of packed cells. Discard supernatant plasma. Wash erythrocytes 3 times by resuspension in 5 mL of cold 0.9% saline. Resuspend packed cells to a volume of 5 mL with 0.9% saline. If more or less than 5 mL of blood is collected, the resuspension should equal the total volume of blood drawn. Record volume of packed cells and total volume of erythrocyte resuspension (red cells & saline).
Minimum Volume:
1 mL washed and resuspended erythrocytes.
Transport:
Frozen on dry ice.
Comments:
The patient should be off medications, if possible, for at least 1 week. If medically inappropriate to discontinue medications, please submit specimen with medication and dosage list.
Rejection Criteria:
Anticoagulants other than heparin; hemolysis; frozen whole blood.
