Specimen Collection Details
Collection: One 7.5 mL serum separator tube (SST). Also acceptable: One 5 mL red top tube.
Handling: Allow to clot, centifuge and separate serum from cells immediately and pour into a plastic vial. Refrigerate until transport. Freeze at -20° C if specimen cannot be assayed within 48 hours. Avoid repeated freeze/thaw cycles.
Standard Volume: 1.5 mL serum.
Minimum Volume: 1 mL serum.
Transport: Refrigerated or frozen on dry ice.
Rejection Criteria: Plasma, gross lipemia or hemolysis, contaminated or heat inactivated specimen; specimen frozen/thawed more than three times.

Neutrophil Cytoplasmic Antibody (ANCA) Screen Result Interpretation, IFA Method:
<1_20 titer
Negative: No significant level of Anti-Neutrophil Cytoplasmic Antibody (ANCA) detected.
≥ 1:20 titer
Positive: Anti-Neutrophil Cytoplasmic Antibody (ANCA) detected.
Samples are screened on ethanol fixed slides which allow differentiation of C and P patterns. Negative results are reported. Positive results are serially diluted and tested on ethanol fixed slides to establish titer and confirm pattern. All P-ANCA patterns are also tested on formalin fixed slides to confirm P-ANCAs by removing Anti-Nuclear Antibody (ANA) receptor sites and maintaining cytoplasmic localization of Myeloperoxidase or other antigens.
Positive ANCA Screen results will reflex to ANCA Titer (69175) and Neutrophil Cytoplasmic Antibody (ANCA) MPO/PR3 Confirmation (69171) if needed. Additional charges will apply.
