Specimen Collection Details
Collection: One 4 mL lavender top tube (EDTA).
Handling: Submit whole blood at ambient temperature. Specimen must arrive at laboratory within 24 hours of collection.
Stability: 24 hours ambient.
Standard Volume: 4 mL whole blood.
Minimum Volume: 1 mL whole blood.
Transport: Ambient.
Comments: The underlying defect in paroxysmal nocturnal hemoglobinuria (PNH) has been traced to a gene mutation which codes for a GPI anchor protein which binds complement-regulating proteins to the red cell surface. Absence of GPI anchored proteins such as CD55 and CD59 is a highly specific indication of presence of the PNH defect.
Rejection Criteria: Clotted, refrigerated or frozen specimen; tubes other than specified; specimen received in laboratory more than 24 hours after collection.
