Specimen Collection Details
Collection: LYMPH NODE: Submit as much tissue as possible.
PERIPHERAL BLOOD: One 4 mL lavender top tube (EDTA).
BONE MARROW: 1-3 mL bone marrow in a 4 mL green top tube (sodium heparin), one 4 mL lavender top tube (EDTA), or one yellow top tube (ACD). Lithium heparin is acceptable but not preferred for flow cytometry testing, and is specifically unacceptable for cytogenetic testing, as it prevents cell division.
BODY FLUID: At least 4 mL fluid preferred. Larger volumes will improve results for fluids with low cell counts.
Handling: LYMPH NODE: Place in ambient temperature RPMI media, available from Client Services, and keep ambient or refrigerated. Submit to the laboratory within 6 hours of collection, or if a delay occurs, mince tissue with scissors to liberate cells and pour into media. If RPMI or other tissue media is not available, sterile saline may be used. Do not allow tissue to dry.
PERIPHERAL BLOOD: Hold ambient or refrigerated and submit within 24 hours.
BONE MARROW/BODY FLUID: Mix bone marrow or body fluid with an equal volume of RPMI media, available from Client Services. Submit to the laboratory within 24 hours.
ALL SPECIMENS: Include relevant history with specimen.
Standard Volume: As much lymph node or body fluid as possible; or 3 mL bone marrow; or 4 mL EDTA whole blood.
Minimum Volume: 0.5 x 0.5 cm lymph node; or 1 mL bone marrow or body fluid; or 4 mL EDTA whole blood.
Transport: Ambient; refrigerated specimen is acceptable.
Rejection Criteria: Poor cell viability; insufficient cells for analysis.
