Specimen Collection Details
Collection: One 7.5 mL serum separator tube (SST). Also acceptable: One 4 mL green top tube (sodium heparin), or one 4 mL lavender top tube (EDTA). Draw following overnight (12 hour) fast. Patient must not consume alcohol for 24 hours prior to blood draw. Protect from light during collection, storage, and transport.
Handling: Allow to clot, centrifuge and separate serum or plasma from cells and pour into a plastic transport tube immediately. Protect from light. VERY LIGHT SENSITIVE. Refrigerate or freeze immediately. Specimen exposed to excessive light may show low results.
Stability: Specimen stable 1 month refrigerated or 1 year frozen. Specimen is unstable ambient.
Standard Volume: 1 mL serum or plasma.
Minimum Volume: 0.3 mL serum or plasma.
Transport: Refrigerated; or if frozen, on dry ice.
Comments: Serum retinol is typically maintained until hepatic stores are almost depleted. Values >0.20 mg/L probably represent adequate liver stores, whereas values < 0.10 mg/L may indicate severe deficiency. In acute hypervitaminosis A, retinol is often >18 mg/L and retinyl palmitate is often >5 mg/L.

Vitamin A supplements in the range of 10,000 IU/day are typically associated with retinyl palmitate <0.10 mg/L. Toxicity can occur when retinol exceeds the capacity of retinol binding protein (RBP); individuals with compromised renal function can retain RBP and may, therefore, have moderate retinol elevations. Drugs which interfere with vitamin A analysis include probucol (Lorelco).
